چکیده

Various anatomically distinct regions from postmortem human brain were analyzed for o-bungarotoxin (a-BuTX) binding and choline acetyltransferase activity. There was a heterogeneous distribution of both cr-BuTX and enzyme activity in the regions studied. Although levels of cY-BuTX binding were lower than those reported in other mammalian brain studies, the relative regional concentration was, in general, similar to that of other mammals. High binding activity was noted in the mammillary body, uncus, colliculi. and cortex. Lowest activity was observed in the caudate nucleus and cerebellum. Choline acetyltransferase activities in parallel assays confirmed prior data in human brain.

The Effect of Swimming Endurance Exercise on Cell Death and Nicotinic Acetylcholine Receptor Gene Expression in Brain of Rat: An Experimental Study of Alzheimer's Disease Model

Background and Objectives: Alzheimerchr('39')s disease (AD) is a neurodegenerative disease which is marked by impaired cholinergic function and decreased nicotinic acetylcholine receptor (nAChRs) density. nAChRs are important mediators of cholinergic signaling in modulation of learning and memory function. In Alzheimer hippocampus is particularly vulnerable to specific degenerative processes an...

Effect of the cholinergic and opioid receptor mechanisms on nicotine-induced analgesia

In this study, we investigated the effect of nicotinic receptor agonists and antagonists on the analgesic response to morphine in the formalin test. In experiments conducted in mice, nicotine produced an early dose-dependent analgesic effect. At a dose of 0.5 mg/kg, mecamylamine, a nicotinic receptor inhibitor, suppressed the analgesic effect induced by 0.1 mg/kg nicotine in both stages of th...

Acetylcholine receptors in the human retina.

Evidence for a population of acetylcholine (ACh) receptors in the human retina is presented. The authors have used the irreversible ligand 3H-propylbenzilylcholine mustard (3H-PrBCM) to label muscarinic receptors. 3H- or 125I-alpha-bungarotoxin (alpha-BTx) was used to label putative nicotinic receptors. Muscarinic receptors are apparently present in the inner plexiform layer of the retina. Auto...

Pharmacological properties of immuno-isolated neuronal nicotinic receptors.

Recently we immunoaffinity-purified an ACh receptor from chicken brain using a monoclonal antibody raised against receptors from fish electric organ (Whiting and Lindstrom, 1986). This neuronal receptor could be affinity-labeled with 3H-bromoacetylcholine, and antisera to it specifically blocked ACh-induced depolarization of chicken ciliary ganglion cells. Here we show that this neuronal ACh re...

Nicotinic receptor abnormalities of Alzheimer's disease: therapeutic implications.

The neuronal nicotinic acetylcholine receptors (nAChRs) in the brain are important for functional processes, including cognitive and memory functions. The nAChRs acting as neuromodulators in communicative processes regulated by different neurotransmitters show a relatively high abundance in the human cortex, with a laminar distribution of the nAChRs of superhigh, high, and low affinity in the h...
